Dr. Giese's research is in the detection and identification
of trace biomolecules such as genotoxins, DNA adducts,
steroids, and bio-active peptides by mass spectrometry. The
work combines analytical chemistry, organic chemistry, and
biochemistry to advance the reagents and techniques available
for sample preparation. Novel tags are developed for
derivatization to increase analyte response and selectivity. A
recent example is the development of a nucleotide exposure
mass profile test which discovers genotoxins in environmental
mixtures. In this test, a DNA nucleotide or strand is used as
bait to catch genotoxic chemicals, giving specific changes in
mass. Because accurate masses are determined, a connection can
be made to corresponding DNA adducts in biological samples
such as human blood lymphocytes. The opportunity to scale up
environmental samples provides a pathway to structural
characterization of unknown genotoxins. Below is a figure
demonstrating the diversity of DNA adducts that can be
detected simultaneously with high sensitivity and similar
response with this technology.
